Yes, we do. Turkish changes with suffixes and sometimes smallest things can change meaning.

Example:

Aylin'de olacağız. (We'll be at Aylin's)

Aylin de gelecek. (Aylin will also come)

I need to correct your sentences though. The question should be: "Nerede yaşıyorsunuz?" (This is for either plural you or formal you. In this case it is formal you question) And the answer is: "Ben ailemle Amerika'da yaşıyorum."
